酸奶在製作過程中要進行髙溫殺菌，目前乳品廠多採用巴氏殺菌，（上海冷庫安裝）即採用95℃的髙溫進行殺菌處理。酸奶滅菌之後，還要進行髙溫發酵（發酵溫度在40℃~43℃之間），發酵之後便要採取冷卻措施。冷卻措施就是讓酸奶發酵後快速冷卻到5℃，因爲酸奶中含有的保加利亞乳杆菌和噬熱鏈球菌，它們的生長溫度分别爲22℃和20℃。如果在規定時間内達不到溫度要求，則會嚴重影響酸奶的菌群數量、營養物質、風味等。
